A non-connexon protein (MIP) is involved in eye lens gap-junction formation.

چکیده
New immunolocalization data put the role of the lens MP26 (MIP) protein in a new perspective. During maturation of lens fibre cells, MIP is found to associate specifically with two structures, gap junctions and cell interlocking processes (known as ball and socket domains). It is significant that the zone in which these associations are most striking is discrete, coinciding with the zone of rapidly enlarging junctional plaques and newly forming ball and socket domains. Observation of domain-specific interactions of MIP with forming gap junctions and ball and socket domains suggests that MIP may be involved in the formation of close membrane appositions. Furthermore, previous ambiguities in the literature over the presence of MIP in gap junctions are clarified by the knowledge that, in situ, MIP associates strongly with gap junctions for only a brief period (with less than about 5% of all lens gap junctions at any one time) during the assembly of junctional plaques.

Dissociation of lens fibre gap junctions releases MP70.
MIP and MP70 are putative gap junction components in the plasma membranes of the mammalian lens fibre cells. We show now that MP70 can be solubilized separately from MIP in mild detergent solutions, and that this treatment results in the dissociation of the fibre gap junctions. Solubilized MP70 was isolated as 16.9 S particles by velocity gradient centrifugation and in the electron microscope h...
متن کاملInteraction of major intrinsic protein (aquaporin-0) with fiber connexins in lens development.
We observed that chick lens-fiber gap-junction-forming proteins, connexin (Cx) 45.6 and Cx56, were associated with an unknown protein, which was then identified as major intrinsic protein (MIP), also known as aquaporin-0 (AQP0), the most abundant membrane protein in lens fibers. A 1063 bp cDNA of chick MIP(AQP0) was identified that encodes a 262 amino acid protein with a predicted molecular wei...
متن کاملReconstitution of native-type noncrystalline lens fiber gap junctions from isolated hemichannels
Gap junctions contain numerous channels that are clustered in apposed membrane patches of adjacent cells. These cell-to-cell channels are formed by pairing of two hemichannels or connexons, and are also referred to as connexon pairs. We have investigated various detergents for their ability to separately solubilize hemichannels or connexon pairs from isolated ovine lens fiber membranes. The sol...
متن کاملImmunocytochemical localization of the main intrinsic polypeptide (MIP) in ultrathin frozen sections of rat lens
The in situ distribution of the 26-kdalton Main Intrinsic Polypeptide (MIP or MP 26), a putative gap junction protein in ocular lens fibers, was defined at the electron microscope level using indirect immunoferritin labeling of ultrathin frozen sections of rat lens. MIP was found distributed throughout the plasma membrane of the lens fiber cell, with no apparent distinction between junctional a...
متن کاملCardiac myocyte gap junctions: evidence for a major connexon protein with an apparent relative molecular mass of 70,000.
It is widely accepted that there is a family of gap junction connexon proteins, their distribution appearing to vary with tissue type and species. In cardiac tissues the major junctional channel component identified is a 43K (K = 10(3) Mr) polypeptide. Using a gap junction isolation protocol in which low temperatures are maintained, and which is detergent-free, we have identified a second gap j...
